[Abstract]

In the 21 century, scientists worked in biomedical areas face a tremendous transformation from genome to proteome. Proteomics is a newly emerging field in the post-genomics era . The development of knowledge and technology in proteomics has provided a new way to study cancer markers.At present there is no serological parameter with high sensitivity and specificity for diagnosing colorectal cancer. This study aimed to establish a serum protein fingerprinting technique coupled with a pattern-matching algorithm to distinguish colorectal cancer from benign colorectal diseases and healthy people. Proteomic spectra were generated by mass spectroscopy (surface-enhanced laser desorption and ionisation). A learning group of spectra derived from analysis of serum which included 31 healthy people, 16 benign colorectal disease, and 73 patients with colorectal cancer was analyzed by an iterative searching algorithm in order to identify a proteomic pattern that completely discriminated cancer from noncancer. The discovered pattern was then used to classify an independent test group of 120 double-blind serum samples: 73 patients with colorectal cancer, 31 healthy people, and 16 benign colorectal diseases. At the key M/Z values 4 467 Da, 8 131 Da, 8 939 Da, 9 192 Da, 9 134 Da, 8 221 Da, 5 928 Da, 8 324 Da, 11 732 Da , protein contents of the three classes in learning group were obviously different by the software analysis. And the classification rule of tree was discovered, and corresponding correct ratio was98.33%(118/120), corresponding sensitivity was 97.26%(71/73) and corresponding specificity was 100%(47/47) ; The test group was examined double-blindly with that rule, corresponding correct ration was 96.77%(116/120), corresponding sensitivity was 95.89%(70/73) and corresponding specificity was 97.87%(46/47). Via comparative proteomics research in the serum of colorectal cancer patients, benign colorectal diseases, and healthy people by SELDI-TOF-MS, colorectal cancer can be quickly and exactly diagnosed by Serum Proteomic Patterns with high sensitivity and specificity.To explore the similarity of biomarkers expression between medium of lovo cells and medium of HT29 cells and lovo cell lysates and serum of colorectal cancer. There were proteins at the M/Z values 2812D?3144D? 3581D?4080D?5198D?9436D?11731D?14892D in the loVo cells suspension. At the M/Z values 2812D?5051D?6098D?6215D?6268D?6980D?7456D?14873D? 14945D?15122D?15180D in the HT29 cells medium. At the M/Z values 6894D? 9521D?9625D?11074D?13778D?14007D in the loVo cells lysates.Therewere similarity of biomarkers expression between lovo cells medium and HT29 cells medium at the M/Z values 2810D?14890D.
